Allyloxycarbonyl (Alloc) is one of the most frequently used alkoxycarbonyl amino protecting groups. It has strong stability to acids and bases, and in its presence, Cbz, Boc and Fmoc can be selectively deprotected.

Introduction of Alloc

The Alloc group can be introduced through Alloc-Cl or Alloc-OSu in organic solvents/Na2CO3, NaHCO3 solution or pyridine.

Removal of Alloc

The removal of Alloc usually requires specific catalytic conditions. ‌Common catalysts are Pd(0) compounds, such as Pd(PPh3)4 or Pd(PPh3)2Cl2. In addition, the selection of a suitable nucleophilic reagent is also important for the removal of Alloc. Commonly used nucleophilic reagents include morpholine, 1,3-diketone, etc.
