Synthesis Methods

Some common methods we use to synthesize bicyclic peptides include:

  • Chemical synthesis. Chemical synthesis is the most common method for creating bicyclic peptides. In this approach, the two cyclic structural elements are typically synthesized separately and then joined together through a linker region. This can be achieved using solid-phase peptide synthesis (SPPS) or solution-phase peptide synthesis.
  • Disulfide bond formation. Bicyclic peptides can also be synthesized through the formation of disulfide bonds. In this method, two linear peptide chains containing cysteine residues are first synthesized. The cysteine residues are then oxidized to form a disulfide bond, resulting in a bicyclic peptide structure.
  • Ligation. Ligation is another method for synthesizing bicyclic peptides, where two linear peptide chains are linked together to form a cyclic structure. This can be achieved using chemical ligation methods such as native chemical ligation (NCL) or thiol-ene click chemistry.
