About Peptoids And Peptides
Peptoids are a class of biomimetic polymers that mimic the structure and function of natural peptides, with side chains attached to the nitrogen atoms of the peptide backbone rather than to α-carbon (as they are in amino acids). In natural peptides, the R groups represent 20 different substitutions of specific amino acids, whereas in peptoids, the choice of R groups can be much wider and potentially unlimited.

Peptoid Synthesis Methods
- Monomer method. This method is very similar to peptide solid-phase synthesis, but before synthesizing peptoids, all protected monomers need to be synthesized. The synthesis process of each protected monomer is tedious and time-consuming.
- Submonomer method. This method does not require α-amino-protected monomers. It is divided into two steps: acylation reaction and substitution reaction. In the acylation reaction, the haloacetic acid is first activated and then reacts with the amine remaining at the end of the previous step. The most common is to activate bromoacetic acid with diisopropylcarbodiimide. In the substitution reaction (the classic SN2 reaction), the amine replaces bromine to form N-substituted glycine. The submonomer synthesis method has great advantages over the monomer synthesis method and has been widely used in the construction of peptoid libraries. We currently synthesize peptoids using the submonomer approach.
