The basic structure of macrolide antibiotics (MALs) has a highly substituted macrolide ring. MALs have high inhibitory activity against gram-positive bacteria and mycoplasma, and are widely used in the treatment of various infectious diseases in food-producing animals. MALs can also be added to animal feed as growth promoters, which may lead to excessive use of MALs.
MALs easily accumulate in animals. Its side effects have been found in the clinic. Common side effects include gastrointestinal reactions, allergic reactions, severe cochlear nerve damage, hearing loss, liver and kidney damage, and drug resistance.
Macrolide antibiotics are mainly metabolized by the liver, through cytochrome P450 biotransformation or other metabolic methods, to produce physiologically active metabolites or inactive metabolites. In addition to the detection of original drugs, the detection of MALs metabolites also needs to be taken seriously. Many metabolites of macrolide drugs retain their active groups and have antibacterial spectrum and antibacterial activity similar to the original drug.
Overuse or unreasonable application of MALs may put consumers at risk. Many countries have also established maximum residue limits for MALs. Food Macrolides Residue Testing Platform

- QuEChERS (Quick, Easy, Cheap, Effective, Rugged, and Safe) method
- Solid-phase extraction
Sample detection technology:
- Enzyme-linked immunoassay (ELISA): This method can only be used for screening, not as a confirmation method.
- Reversed-phase high performance liquid chromatography: detection limit is 0.016-0.041mg/kg, quantification limit is 0.053-0.135 mg/kg.
- Liquid chromatography-electrospray mass spectrometry (LC / ESI-MS / MS): Data acquisition under MS / MS is achieved by multi-reaction monitoring (MRM) of two or three fragment ion transitions. It is more sensitive than liquid chromatography. The detection limit is less than 1.0μg/kg, and the quantification limit is 0.005-0.030mg/kg.
