Food Amino Acid Testing Platform

  • Ninhydrin post-column derivatization method

The cation exchange resin is used as the stationary phase and the acid buffer is used as the mobile phase. The sample undergoes high-efficiency cation exchange chromatography. Ninhydrin is added to the post-column effluent to indirectly detect derivatives with visible light absorption generated by amino acids. Photoamine and o-phthalaldehyde can replace ninhydrin to increase the sensitivity by 3 orders of magnitude. This method is not suitable for proline detection.

  • Pre-column derivatization reversed-phase high performance liquid chromatography

Choose a suitable derivatizing agent and quantitatively react with each amino acid to produce a specific product with a certain stability, and then analyze the product by high performance liquid chromatography to reverse the amino acid.

This method has the advantages of fast speed (all analysis results can be obtained in 1 min), no pollution (no chemical reagent discharge), no sample damage, good experiment reproducibility (no-man error), and high accuracy. It can detect free amino acids as well as bound amino acids.

Not suitable for small batch sample analysis. The detection range of this technology cannot completely cover twenty standard amino acids. The scope of coverage depends on the nature of the solution during sample pretreatment and the nature of the amino acid itself (mostly depending on the side chain).

Acceptable Samples: Nearly all raw and processed foods or ingredients.
