In studies on neuropathic pain, it effectively alleviates symptoms in chronic constriction injury (CCI) rats by inhibiting the activation of the p38 MAPK and NF-κB signaling pathways in the spinal cord. Additionally, saikosaponin A modulates neurological activity by inhibiting NMDA receptor currents, persistent sodium currents, and the production of pro-inflammatory cytokines such as TNF-α and IL-1β, along with reducing caspase-1 activity. These properties position it as a promising candidate for further research into pain management and neuroprotection.

Saikosaponin D

Saikosaponin D is a prominent bioactive compound found in Bupleurum chinense, extensively researched for its medicinal applications. As a calcium mobilizing agent and SERCA inhibitor, it also acts as an agonist of the glucocorticoid receptor (GR), exhibiting significant anti-cancer, anti-inflammatory, and neuroprotective properties. Saikosaponin D has been shown to protect against acetaminophen-induced liver toxicity by suppressing NF-κB and STAT3 signaling pathways, leading to reduced expression of iNOS, COX-2, and pro-inflammatory cytokines. Additionally, it inhibits apoptosis and mitigates the mitochondrial translocation of GR in corticosterone-treated PC12 cells, highlighting its neuroprotective effects. Saikosaponin D also improves depressive-like behaviors in various models of depression, further emphasizing its potential in neuropsychiatric treatments.

Saikosaponin C

Saikosaponin C is a naturally occurring triterpenoid saponin primarily found in plants of the Bupleurum genus. While it has not been studied as extensively as saikosaponins A and D, emerging research suggests that saikosaponin C holds promising pharmacological potential. Notably, it exhibits anti-HBV (hepatitis B virus) activity and shows potential for promoting therapeutic angiogenesis, although it is not considered suitable for cancer treatments. Additionally, saikosaponin C may serve as a novel therapeutic option for neurodegenerative conditions such as Alzheimer's disease.
