Discovery. The term nanozymes was first coined by Pasquato, Scrimin, and co-workers in 2004 to describe the transphosphorylation reactivity of triazacyclononane-functionalized gold nanoparticles (NPs). In 2007, Yan's research group first discovered that Fe3O4 magnetic nanoparticles have catalytic activity similar to natural horseradish peroxidase. They demonstrated that Fe3O4 NPs could catalyze the oxidation of peroxidase substrates (i.e., 3,3,5,5-tetramethylbenzidine (TMB), diazoaminobenzene (DAB), and o-phenylenediamine (OPD)) in the presence of H2O2, resulting in a colorimetric reaction, showing the peroxidase-like activity of Fe3O4 NPs toward typical peroxidase substrates[1]. Since then, nanozymes have specifically referred to nanomaterials with intrinsic enzyme-like properties.
Development. As a new type of simulated enzyme, nanozymes not only have the unique physical properties of nanomaterials, but also have efficient catalytic functions similar to natural enzymes. Since their discovery, research on nanozymes has risen rapidly. More and more nanomaterials or nanocomposites have been found to have enzyme-simulating properties, such as prussian blue NPs, ceria NPs, cobalt oxide NPs, noble metal NPs of Au, Ag, Pt, Pd, Ru, etc., and bimetallic PtRu NPs, fullerene and its derivatives, graphene oxide, carbon nanotubes, etc. With the continuous deepening of research, the types of nanozymes have been continuously enriched, and their application fields are also constantly expanding, including biosensing, environmental treatment, disease treatment, etc. The development of nanozymes is a process of continuous exploration and innovation, and their research and application prospects are very broad. The priority development direction in the future will focus on exploring new activities and new materials of nanozymes, studying the catalytic behavior of nanozymes, optimizing the design of nanozymes, and studying the immunocompatibility of nanozymes in vivo.
