Cyclodextrins are cyclic oligosaccharides obtained from the biodegradation of starch, which contain glucopyranose units linked to each other by α-(1,4) bonds. Due to the chair conformation of the glucopyranose units, the cyclodextrin assumes a torus-like shape (truncated cone) with primary hydroxyl groups located on the narrow edge of the cone and secondary hydroxyl groups located on the wider edge of the cone. There are skeletal carbons with hydrogen atoms and oxygen bridges within the cyclodextrin molecule, resulting in the hydrophilic outer surface and lipophilic central cavity of the molecule [1],[2]. The torus-like shape of cyclodextrin is shown in Fig. 1.

Since the late 1970s, cyclodextrins as versatile supramolecular hosts have attracted considerable attention for scientific exploration and have become the most important research topic. Native cyclodextrins, cyclodextrin derivatives, and cyclodextrin inclusion complexes exhibit extensive commercial applications, such as pharmaceuticals, biomedical, cosmetics, food, agricultural, textile, and pharmaceutical applications.

Advantages of Cyclodextrins

Cyclodextrins have become a popular choice in pharmaceuticals and many fields due to their nontoxicity, low cost, safety, and easy availability, especially their ability to form inclusion complexes with guest molecules. The following are the main advantages of cyclodextrins [1]:

  • Enhance solubility: Cyclodextrins can significantly enhance the apparent water solubility of molecules by forming dynamic, non-covalent, water-soluble inclusion complexes with poorly soluble or insoluble compounds.
  • Improve stability: The formation of a complex between cyclodextrin and unstable compounds can significantly improve the stability of the guest molecules, which through slowing down the hydrolysis, oxidation, photolysis, dehydration and isomerization of active molecules.
  • Improve safety: Cyclodextrins can reduce the irritation and toxicity caused by guest molecules in the form of host-guest inclusion complexes to improve the safety of guest molecules. For example, cyclodextrin-drug inclusion complexes can reduce drug-related side effects and irritation by protecting biological membranes from direct contact with drugs.
