Cyclodextrins (CDs) are a class of cyclic oligosaccharides with d-glucopyranoside units linked by glycosidic bonds. The chemical properties and types of CDs were first described in 1891 [1]. Due to the structure of a hydrophilic outer surface and a lipophilic central cavity, CDs are capable of forming water-soluble compounds with a variety of drug molecules.

As understanding and research on CDs and their derivatives have deepened over the years, their applications in the pharmaceutical and biomedical fields have attracted more attention from scientific researchers, and relevant research reports are growing daily. CDs have been proven to work as complexing agents to increase the water solubility of poorly soluble drugs [2], as well as improve the stability [3] and bioavailability of drug molecules. In addition, CDs can also reduce or eliminate the unpleasant odor of drugs, and reduce the toxic side effects and irritation of drugs [4].

Pharmaceutical and Biomedical Applications

Due to the various characteristics and advantages exhibited by CDs, their applications have been widely explored in the pharmaceutical and biomedical fields. A comprehensive introduction to the applications of CDs is described below, including drug extraction, drug separation, drug delivery, drug discovery and biomedical engineering.

Drug Extraction

CDs can be used as auxiliary extraction agents for Chinese materia medica (CMM) extraction, resulting in complete one-step extraction and inclusion. Extraction methods include solvent extraction, microwave assisted extraction, ultrasonic assisted extraction, and mechanochemical assisted extraction.

Drug Separation

CDs can be used as chiral mobile phase additives and chiral stationary phase for the separation of chiral drugs. Different separation methods include high-performance liquid chromatography (HPLC), gas chromatography (GC), mass spectrometry (MS), and supercritical fluid chromatography (SFC).

Drug Delivery

CDs are suitable as pharmaceutical excipients for use in various drug delivery systems, including oral drug delivery system, rectal drug delivery system, nasal drug delivery system, and transdermal drug delivery system.

Drug Discovery

New drugs can be developed and discovered using CD molecules because certain CDs exhibit pharmaceutical activities, such as antiviral and antiparasitic activity.

Biomedical Engineering

CDs can be used in biomedical engineering, and specific applications include biomimetic corneal transplantation, bioimaging, biosensors, medical device coating, and tissue engineering.
