With continued research and development into cyclodextrins (CDs), over 1500 different CD derivatives have been described in the literature. These modified CDs still retain a cavity structure that can encapsulate various guest molecules, which exhibit better complexing capabilities than the parent CDs. To better understand the physical and chemical properties of CD derivatives for targeted applications, CD analysis is essential. And the complexity of CD derivatization requires more comprehensive analytical methods.

Alfa Chemistry has been exploring and developing CDs for many years. We have a modern analytical testing laboratory staffed by professional analytical experts and equipped with advanced testing equipment. With our comprehensive CD analysis services, customers are able to make sure products meet their requirements.

Main Analytical Techniques

Analytical TechniquesDescription
High-performance liquid chromatography (HPLC)HPLC is the best way to analyze the small differences of heterogeneous derivatives. It can be used for purity determination of CD samples, qualitative and quantitative determination of CD components in formulations, and impurity separation for CD samples. HPLC can be applied to analyze Sugammadex samples.
*Sugammadex is a modified single isomer γ-cyclodextrin indicated for rapid reversal of neuromuscular blockade induced by rocuronium bromide and vecuronium bromide.
Gas chromatography (GC)GC is an analytical method for testing the purity of a substance, or separating the different components of a mixture. It can be used to determine the content of volatile organic components, residual solvents, volatile active ingredients and other components in CD-guest inclusion complexes, and evaluate the interaction strength between volatile guest molecules and CD molecules.
Nuclear magnetic resonance spectroscopy (NMR)NMR is a spectroscopic technique that determines molecular structure by providing information such as the functional groups present in a sample. It can be used to identify CDs' structure, determine the interaction strength between active pharmaceutical ingredient (API) and CDs for CD-API inclusion complexes, determine the average degree of substitution for modified CDs, and assess the stoichiometry and complex structure of CD samples.
Capillary electrophoresis (CE)CE is an analytical technique that separates molecules and ions based on their mobility with the use of an applied voltage. It can be used to determine the interaction strength between API and CDs for CD-API inclusion complexes, determine the residual ionic compounds for CD samples, and determine the average degree of substitution of charged CDs.
Mass spectroscopic (MS)

MS is an analytical tool for measuring the mass-to-charge ratio (m/z) of molecules in a sample and is suitable for use with a variety of analytical methods to obtain better analytical results.

  • Inductively coupled plasma mass spectrometry (ICP-MS) can be used to measure elemental impurities at trace levels in CD samples
  • Matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) can be used to determine the molecular weight of CDs and identify the distribution of the degree of substitution for CD polymers.
  • Electron spray ionization-mass spectrometry (ESI-MS) can be applied to determine molecular weights of novel CD derivatives.
